Pharmaceutical properties
Prednisolone is a dehydrogenated analogue of hydrocortisone. The antiallergic, anti-inflammatory, antiexudative and antipruritic effect with external use of the prednisolone ointment causes the decrease in the formation, release and activity of inflammatory mediators (kinin, histamine, lysosomal enzymes, prostaglandins). Cell migration to the epicenter of inflammation is also suppressed, vasodilation and increased vascular permeability that occurs in the focus of inflammation are reduced. The decrease in exudation is due to the vasoconstrictor action of the drug "Prednisolone". The instruction indicates that the drug causes an immunosuppressive effect, inhibiting the action of complex antigen-antibody formations that accumulate on the walls of blood vessels and cause allergic vasculitis. The drug inhibits the action of macrophages, cytokines, target cells, which cause the development of a reaction of the type of allergic contagious dermatitis. In this case, sensitized T-lymphocytes and macrophages cannot gain access to target cells.
Pharmacokinetics
When applying the ointment "Prednisolone" is absorbed and the active substance is in the bloodstream. In plasma, a significant portion of prednisolone is associated with albumin and transcortin. Biotransformation of prednisone occurs by oxidation, mainly in the liver, where glucuronidation or sulfatization of oxidized forms occurs. In the form of metabolites, it is excreted from the body with urine and feces, partly in an unchanged state. Prednisolone passes through the placental barrier, in small concentrations it can be found in breast milk of a woman.
Prednisone: indications
The drug is used in cases of manifestation:
- allergic and contact dermatitis,
- eczema
- atopic dermatitis,
- seborrheic dermatitis,
- psoriasis.
Prednisone: instructions
The ointment is applied to the lesion in an even layer 1-3 times / day. The duration of treatment is determined by the nature of the disease and usually lasts 8-14 days. When treating children with ointment, the treatment period must be reduced to 3-7 days, as the child’s body is more sensitive to the drug “Prednisolone”. The instruction recommends abandoning warming, fixing and occlusive dressings.
Application features
The duration of treatment with ointment should not be more than 14 days in order to prevent possible side effects from the use of the drug "Prednisolone". The instruction prescribes treatment only under medical supervision.
Side effects
Usually the drug is well tolerated. Rarely, especially with prolonged use, itching, burning sensation, and erythema are possible. In some patients, side effects can occur in the form of folliculitis, hypertrichosis, perioral dermatitis, allergic reactions to the components of the drug. The above symptoms do not require treatment and are quickly eliminated when the drug is discontinued. It should be remembered that with the application of the ointment on damaged skin and with prolonged use (especially with lesions of large areas of the skin), systemic side effects may occur.
Interaction with other medicines
Prednisone has the property of enhancing the anticoagulant effect of anticoagulants. The possibility of bleeding increases while taking salicylates. The likelihood of electrolyte metabolic disturbances increases while diuretics are prescribed. The risk of intoxication is increased when using cardiac glycosides. The hypoglycemic effect of antidiabetic agents is reduced and the antibacterial activity of rifampicin is reduced. It is not recommended to be used simultaneously with antacids.
Contraindications
Treatment with ointment "Prednisolone" instruction prohibits with:
- hypersensitivity;
- bacterial, viral, fungal skin lesions;
- tuberculosis, syphilis, skin tumors, pregnancy, acne vulgaris and rosacea.
Overdose
Cases of overdose are unknown.
Storage conditions
Store according to list B, at a temperature of 5-15º. Shelf life is 2 years. Do not use after expiration date .
